Abstract:
Two species of flatfish from southern California were evaluated for exposure to polynuclear aromatic hydrocarbons (PAHs). California halibut (Paralichthys californicus) were collected from eight bays and harbors (Ventura Harbor, Channel Islands Harbor, Marina del Rey, King Harbor, Alamitos Bay, Long Beach Harbor, Newport Bay, San Diego Bay), and a reference site off Camp Pendleton. Pacific sanddab (Citharichthys sordidus) were collected near four of the Channel Islands (San Miguel Isle, Santa Cruz Isle, Anacapa Isle, Santa Barbara Isle). PAH metabolites in fish bile were characterized using a semi- quantitative technique that measures fluorescent aromatic compounds (FACs) in fish bile. The concentration of FACs was elevated in most fish from bays and harbors, compared to reference fish, with average concentrations ranging from three to ten times greater than reference fish. Among the bays and harbors, average FAC concentrations varied by a factor of three. There was no evidence of elevated PAH exposure in the Pacific sanddabs from the Channel Islands.
Purpose:
Many PAHs are toxic and exposure to this group of compounds has been identified as a risk factor influencing the development of liver lesions in fish. However, exposure to PAHs cannot be assessed by conventional tissue analyses because these compounds are rapidly metabolized by the liver and secreted into the bile. One biomarker that has been used to quantify the exposure to PAHs in fish is fluorescent aromatic compounds (FACs) in fish bile. This project was conducted to characterize the PAH exposure in fish from environments contaminated with PAHs.

California halibut (Paralichthys californicus) were collected from eight bays and marinas (Ventura Harbor, Channel Islands Harbor, Marina del Rey, King Harbor, Alamitos Bay, Long Beach Harbor, Newport Bay, San Diego Bay) between July 31 and September 2, 1998. Additional California halibut were collected from a reference site off Camp Pendleton on August 23, 1999. Pacific sanddab (Citharichthys sordidus) were collected near four of the Channel Islands (San Miguel Isle, Santa Cruz Isle, Anacapa Isle, Santa Barbara Isle) between July 28 and September 15, 1998. All fish were captured using an otter trawl. Livers and gall bladders were dissected from the fish at sea and frozen on dry ice or in liquid nitrogen. All tissues were stored in the laboratory at -80C until they were analyzed. Bile from each gall bladder was diluted 1:1 with 50% MeOH, then centrifuged to remove debris that would interfere with analysis. Concentrations of FACs were determined using reverse-phase high performance liquid chromatography (HPLC) and fluorescence detection. The temperature of the reverse-phase column was maintained at 50C using a column heater. Five l of the diluted bile supernatant was injected onto the HPLC. Fluorescence of the PAH metabolites was measured at the excitation/emission wavelength pair (380/430 nm) appropriate for PAHs with similar structure as benzo[a]pyrene and its metabolites. The higher molecular weight PAHs (products of combustion) and their metabolites tend to fluoresce at the same wavelength, and are semi- quantitated collectively as BaP equivalents. Fluorescent values were converted to ng BaP equivalents using a BaP standard curve. Quality assurance procedures included measurement of a California halibut bile composite, a BaP standard, and a 50% MeOH blank at the beginning of each set of analyses. The bile composite was reanalyzed after every seven samples. The concentration of bile proteins was measured using the BioRad DC protein assay, with bovine serum albumin as the protein standard. Protein normalization of the FAC data generally increased the variability among fish from each station, and among each marina or harbor. Protein normalization is supposed to help reduce the variability among FAC measurements, which was not the case with this data. Therefore it is advised that protein normalization should not be conducted with this data.

Process_Description:
Each field crew or laboratory that generated data entered it into their own data management system and subjected it to their internal QA/QC procedures. Recommended QA procedures included double entry of data and range checks. Data was then reformatted following the Standard Data Transfer Protocol (SDTP) and submitted to the Information Management Officer (IMO) in comma- delimited, ASCII format. Standardized data files were submitted to the IMO by diskette, e-mail, or FTP. The IMO then initiated a series of error checks to ensure the data: 1) were within specified ranges appropriate to each parameter measured, 2) contained all required fields, 3) had encoded valid values from constrained look-up lists where specified, and 4) were in the correct format (text in text fields, values in numeric fields, etc.). After all data were submitted, error checked, and corrected, the IMO certified that the file was consistent with the SDTP format requirements and complete. The IMO then notified the chairperson of the Technical Committee responsible for the data that it was ready for technical review. The IMO distributed the data to the chairperson as a comma-delimited ASCII file in the SDTP format. The Technical Committee Chair (TCC) and the Technical Committee reviewed the data with respect to scientific content. This review involved plotting of data and examining interrelationships among individual parameter responses and addressed more extensive data quality issues than can be accomplished by range checking alone. All corrections to the data were made by the IMO. Prior to making any changes, the IMO documented the changes and received (written or electronic) concurrence from the organization that originated the data. The IMO only make changes in the centralized data base; originating organizations were responsible for making corresponding changes in their own internal data storage systems. All changes to the data were documented in a computerized file available to all data users.
